CymR (cymbal repressor) is a bacterial transcriptional repressor from Pseudomonas putida that binds cumate operator (CuO) sequences to silence downstream gene expression. In cumate-inducible expression systems, CymR constitutively represses target gene transcription until cumate is added, relieving repression and enabling tightly controlled, dose-dependent gene expression with minimal background activity.

Experimentally Validated
Each lot undergoes quality control testing to confirm stable transgene expression. Cells are validated through a minimum of 30 serial passages to ensure long-term expression stability, and cryopreserved stocks are tested for viability retention up to 5 years at −80°C. Mycoplasma screening is performed on every lot to confirm negative status.
Usage Notes
Thaw the cryopreserved vial rapidly in a 37°C water bath until a small ice crystal remains. Transfer cells to a pre-warmed culture vessel containing complete growth medium (DMEM supplemented with 10% FBS and appropriate antibiotics). Allow cells to attach for 12–24 hours before changing medium. Maintain selection pressure with zeocin at the recommended concentration (300 µg/mL) to preserve transgene expression.
Culture at 37°C in a humidified atmosphere with 5% CO₂. Passage cells at 70–90% confluency using standard trypsinization protocols. Do not exceed passage 20 for optimal transgene expression stability.
