Scientific Background

mCherry is a monomeric red fluorescent protein derived from Discosoma sp. DsRed, offering bright red fluorescence (excitation 587 nm, emission 610 nm) with excellent photostability. As a spectrally distinct reporter from GFP-family proteins, mCherry enables multicolor imaging experiments, cell tracking, and serves as a reliable fluorescent marker in mammalian cell systems.

Product Description

Stable Cell Line

HEK293T-mCherry is a stable cell line generated by transducing HEK293T cells with the LV-EF1a-mCherry-IRES-Puro lentiviral vector (SKU: BHV22000010), followed by puromycin selection. The constitutive EF1a promoter drives robust mCherry expression, enabling fluorescence-based visualization, imaging, assay development, and experimental optimization. The puromycin-resistance cassette facilitates antibiotic selection and maintenance of a consistently mCherry-positive cell population. The product is supplied cryopreserved at 3 × 10⁶ cells per vial.

QC & Validation

Experimentally Validated

Each lot undergoes quality control testing to confirm stable transgene expression. Cells are validated through a minimum of 30 serial passages to ensure long-term expression stability, and cryopreserved stocks are tested for viability retention up to 5 years at −80°C. Mycoplasma screening is performed on every lot to confirm negative status.

Usage Notes

Thaw the cryopreserved vial rapidly in a 37°C water bath until a small ice crystal remains. Transfer cells to a pre-warmed culture vessel containing complete growth medium (DMEM supplemented with 10% FBS and appropriate antibiotics). Allow cells to attach for 12–24 hours before changing medium. Maintain selection pressure with puromycin at the recommended concentration (1.0 µg/mL) to preserve transgene expression.

Culture at 37°C in a humidified atmosphere with 5% CO₂. Passage cells at 70–90% confluency using standard trypsinization protocols. Do not exceed passage 20 for optimal transgene expression stability.

Safety & Handling

Safety Notice: This product contains genetically modified cells produced using lentiviral transduction. Handle under BSL-2 conditions. Follow institutional biosafety guidelines for work with lentiviral-transduced cell lines. Wear appropriate PPE including gloves, lab coat, and eye protection. Decontaminate all materials that contact cells with 10% bleach or approved disinfectant before disposal.

Related Products

Q.What is the host/parental cell line for this product?
A.This stable cell line is derived from HEK293T cells, a highly transfectable human embryonic kidney cell line widely used in biomedical research. HEK293T cells contain the SV40 large T antigen for enhanced episomal replication of plasmids containing the SV40 origin of replication.
Q.What biosafety level is required?
A.BSL-2 containment is required for handling this lentiviral-transduced cell line. Follow your institution's Institutional Biosafety Committee (IBC) guidelines for work with genetically modified organisms produced using lentiviral vectors.
Q.Has this cell line been tested for mycoplasma?
A.Yes, each lot is tested for mycoplasma contamination and confirmed negative before release. Routine mycoplasma testing during culture is recommended as standard practice.
Q.What selection marker concentration should I use?
A.Puromycin at 1.0 µg/mL is the recommended maintenance concentration for this cell line. A kill curve on the parental HEK293T cells may be performed to optimize selection stringency for your specific culture conditions.
Q.How should I store and recover the cells?
A.Store cryopreserved vials in liquid nitrogen vapor phase. For recovery, thaw rapidly in a 37°C water bath, transfer to pre-warmed complete medium (DMEM + 10% FBS), and allow cells to attach for 12–24 hours before changing medium. Do not store long-term at -20°C or -80°C.
Q.What are the excitation and emission wavelengths for mCherry?
A.mCherry has an excitation maximum at 587 nm and emission maximum at 610 nm. Use Texas Red or mCherry-specific filter sets for fluorescence microscopy. mCherry is spectrally compatible with GFP for dual-color imaging experiments.
