Vector Overview

GCaMP is a genetically encoded calcium indicator (GECI) consisting of a circularly permuted green fluorescent protein (GFP), calmodulin (CaM), and the M13 peptide derived from myosin light chain kinase. Upon binding Ca²⁺, GCaMP undergoes a conformational change that enhances GFP fluorescence, enabling real-time monitoring of intracellular calcium dynamics associated with neuronal activity, receptor signaling, and other Ca²⁺-dependent cellular processes. Because GECIs are genetically encoded, they can be targeted to specific cell types and support long-term imaging in living systems. GCaMP6 is an ultrasensitive generation of GCaMP with substantially improved signal-to-noise ratio and sensitivity for detecting cytosolic Ca²⁺ transients. Different GCaMP6 variants exhibit distinct kinetic properties, including fast (GCaMP6f), medium (GCaMP6m), and slow (GCaMP6s) fluorescence decay rates, allowing selection based on experimental requirements. LV-CAG-GCaMP6s-IRES-Puro is a pre-made lentiviral vector that expresses the slow-kinetics GCaMP6s variant under the control of the strong, ubiquitous CAG promoter, along with a puromycin resistance gene for antibiotic selection. Supplied in a ready-to-use format for efficient gene delivery in vitro or in vivo.
