Vector Overview

LV-CAG-tdTOMATO is a pre-made recombinant lentiviral vector designed for robust and stable expression of the red fluorescent reporter protein tdTomato under the control of the CAG promoter (also known as the CBA promoter). The CAG promoter combines the cytomegalovirus (CMV) immediate-early enhancer with the chicken β-actin promoter, enabling strong and constitutive transgene expression across a broad range of mammalian cell types. tdTomato is a bright red fluorescent protein with excellent photostability and high quantum yield, making it well suited for monitoring transduction efficiency, tracking cells, visualizing gene expression, and performing in vitro and in vivo imaging studies. Following transduction, the lentiviral vector integrates into the host cell genome, enabling long-term and stable expression of tdTomato in both dividing and non-dividing cells. This ready-to-use lentiviral preparation is suitable for applications including cell labeling, lineage tracing, cell tracking, transduction optimization, fluorescence microscopy, flow cytometry, and the generation of stable fluorescent cell lines.

Q.Does this vector include an antibiotic selection marker?
A.No. Transduced cells are identified by the tdTomato reporter — by microscopy, FACS or the matching bulk readout — rather than by drug selection.
Q.Is expression constitutive or regulated?
A.Expression of the cassette is constitutive under the CAG promoter: no inducer or driver line is needed for the vector itself, and expression persists in daughter cells after integration.
Q.How should the vector be stored and handled?
A.Store at -80°C; avoid repeated freeze-thaw cycles. Ships on dry ice.
