Vector Overview
LV-EF1a-CymR-IRES-Puro is a pre-made, replication-deficient lentiviral vector designed for the stable delivery and expression of the CymR repressor protein in mammalian cells. CymR is a regulatory protein derived from Pseudomonas putida that specifically binds to the cumate operator (CuO), enabling tight and reversible transcriptional control in cumate-inducible gene expression systems. In this construct, CymR expression is driven by the human EF1a promoter, providing strong and constitutive expression across a wide range of dividing and non-dividing cell types. The vector also includes a puromycin resistance gene (Puro) linked via an internal ribosome entry site (IRES), allowing efficient antibiotic selection and the generation of stable cell lines following transduction. Packaged using a third-generation lentiviral system, LV-EF1a-CymR-IRES-Puro integrates efficiently into the host genome, enabling long-term and heritable expression of CymR. This vector is particularly well suited for establishing CymR-expressing “repressor” cell lines, which can be paired with CuO-regulated constructs to achieve tunable and inducible gene expression.
