Vector Overview

LV-EF1a-CymR-PGK-Zeo is a pre-made, replication-deficient lentiviral vector designed for stable delivery and expression of the CymR repressor protein in mammalian cells. CymR is a regulatory protein derived from Pseudomonas putida that specifically binds to the cumate operator (CuO), enabling tight transcriptional control in cumate-inducible gene expression systems. In this construct, CymR expression is driven by the human EF1a promoter, which provides strong, constitutive expression across a broad range of dividing and non-dividing cell types. The vector also contains a zeocin resistance gene (Zeo) under the control of the PGK promoter, allowing efficient antibiotic selection and generation of stable cell lines following transduction. Packaged in a third-generation lentiviral system, LV-EF1a-CymR-PGK-Zeo efficiently integrates into the host genome, enabling long-term and heritable expression of CymR. This vector is particularly useful for establishing CymR-expressing “repressor” cell lines that can subsequently be used with CuO-regulated expression constructs for tunable, inducible gene expression.
