Vector Overview
FLP recombinase, originally identified in Saccharomyces cerevisiae, is a site-specific recombinase (SSR) belonging to the λ integrase family that recognizes a distinct 34 bp FRT sequence. The FLP/FRT system has been widely used to generate conditional knockout mice. However, early applications of FLP in mammalian cells revealed inefficient recombination activity due to the thermoinstability of the FLP protein at 37 °C. Subsequent screening for thermostable variants led to the development of FLPe, which exhibits approximately a fourfold increase in recombination efficiency at physiological temperature. Despite this improvement, FLPe still displays relatively low recombination efficiency in mammalian cells because of its non-mammalian origin. FLPo is a mouse codon-optimized version of FLPe that significantly enhances FRT-mediated recombination efficiency both in vitro and in vivo in mice. The LV-EF1a-FLPo-Puro pre-made lentivirus expresses FLPo recombinase under the control of the EF1a promoter, with puromycin resistance co-expressed via an IRES cassette. This lentivirus is supplied in a ready-to-use format.
