Vector Overview
LV-EF1a-hGluc-Puro is a pre-made lentivirus that overexpresses codon-optimized, humanized Gaussia luciferase (hGluc) under the control of the EF1a promoter, with puromycin resistance co-expressed via an IRES cassette. The humanized Gaussia luciferase (hGluc) has been validated as non-toxic and is naturally secreted following overexpression in mammalian cells. Compared to humanized firefly (hFLuc) and Renilla (hRLuc) luciferases expressed under similar conditions, hGluc produces over 1,000-fold higher bioluminescent signal intensity in live cells and more than 100-fold higher signal intensity from viable cells alone (excluding secreted luciferase) or from cell lysates. In vivo, hGluc generates approximately 200-fold higher signal intensity than hRLuc and exhibits signal levels comparable to hFLuc under standard imaging conditions. Due to its high signal intensity, natural secretion, and ATP-independent activity, Gaussia luciferase provides a highly sensitive and versatile reporter for monitoring gene delivery and other biological events in real time, both in cultured cells and in vivo. This lentivirus is supplied in a ready-to-use format.
