Vector Overview
LV-EF1a-SpCas9(D10A)-Puro is a pre-made lentiviral vector that expresses the Cas9 D10A nickase mutant derived from Streptococcus pyogenes (SpCas9[D10A]) under the control of the human elongation factor-1 alpha (EF1a) promoter. The vector also carries a puromycin resistance gene linked via an internal ribosome entry site (IRES) cassette, enabling antibiotic selection of transduced cells. The D10A mutation converts wild-type Cas9 into a DNA nickase that cleaves only a single DNA strand. When paired guide RNAs are designed to target opposite DNA strands in close proximity, SpCas9(D10A) generates highly specific double-strand breaks while substantially reducing off-target genome editing compared to wild-type Cas9. As a result, this system is widely used for precise genome engineering, gene knockout studies, and CRISPR-based functional genomics applications. The EF1a promoter drives robust and stable transgene expression across a wide variety of mammalian cell types, including primary cells and stem cells. Puromycin selection enables efficient enrichment of transduced cell populations for downstream experimental applications. This lentiviral vector is supplied in a ready-to-use format for both in vitro and in vivo research applications.
