Lentiviral Vector

Vector Overview

LV-UBC-mCherry-GFP-LC3 is a pre-made lentiviral vector designed to overexpress an mCherry-GFP-LC3 fusion protein under the control of the UBC promoter in mammalian cells. This vector serves as a powerful tool for monitoring autophagic flux: autophagosomes are visualized by the presence of both GFP and mCherry fluorescence, while autolysosomes—formed when autophagosomes fuse with lysosomes—display mCherry fluorescence only, as GFP fluorescence is quenched in the acidic lysosomal environment. This dual-color system enables real-time tracking of autophagy dynamics in live cells. The lentivirus is supplied in a ready-to-use format for efficient transduction and stable expression.

Biosafety

LV-UBC-mCherry-GFP-LC3 is a pre-packaged lentiviral vector classified as BSL-2. Handle in a certified biosafety cabinet under institutional BSL-2 practices, decontaminate all contact surfaces and waste, and follow local regulations for viral vector work.

Safety & Handling

Store at -80°C; avoid repeated freeze-thaw cycles. Ships on dry ice. Supplied in PBS. For research use only; not for diagnostic or therapeutic use.

Q.What biosafety level is required for LV-UBC-mCherry-GFP-LC3?
A.LV-UBC-mCherry-GFP-LC3 is a lentiviral vector handled at BSL-2. Work in a certified biosafety cabinet, wear appropriate PPE and decontaminate all waste; institutional biosafety approval is typically required.
Q.What titer is supplied and how should MOI be chosen?
A.The vector is supplied at >1E+9 TU/mL functional titer, stated as a minimum. Calculate multiplicity of infection (MOI) from this value and the number of target cells, and optimize MOI empirically for each cell type, since transduction efficiency varies between cell lines and primary cells.
Q.Does this vector include an antibiotic selection marker?
A.No. Transduced cells are identified by the mCherry-GFP-LC3 reporter — by microscopy, FACS or the matching bulk readout — rather than by drug selection.
Q.Is expression constitutive or regulated?
A.Expression of the cassette is constitutive under the UBC promoter: no inducer or driver line is needed for the vector itself, and expression persists in daughter cells after integration.
Q.How should the vector be stored and handled?
A.Store at -80°C; avoid repeated freeze-thaw cycles. Ships on dry ice.

Common customization requests

  • Insert / payload: replace the gene/sequence, swap to a different isoform, add mutations, or optimize cloning features.
  • Expression design: change promoter (e.g., CMV/EF1α/PGK), add enhancers, or adjust regulatory elements.
  • Reporters: add/swap GFP/RFP/mCherry/luciferase (single or dual reporters where applicable).
  • Selection markers: add/swap puromycin/blasticidin/neomycin or fluorescent selection options.
  • Vector format: switch between OE, shRNA, CRISPR (sgRNA/Cas systems), or control vectors (where supported).

Add-ons you can request

  • Control viruses: empty vector, non-targeting shRNA, reporter-only controls, or matched backbone controls.
