Bacterial DNA topoisomerase II (gyrase) is a type II topoisomerase that converts relaxed circular DNA into negatively supercoiled DNA. Type II topoisomerases cut and rejoin both DNA strands to change DNA topology.
Some inhibitors, such as fluoroquinolones, block the rejoining step; the enzyme then stays covalently linked to cut DNA in a cleavage complex. Ciprofloxacin and novobiocin both inhibit gyrase, but only ciprofloxacin produces cleavage complexes.

Use With
This enzyme is used in the following ProFoldin assay kits:
- 96-Well S. aureus Gyrase DNA Decatenation Assay Kit Plus (BHT16300066) — complete kit: already includes this enzyme
- S. aureus Gyrase ATPase Assay Kit Plus (BHT16300074) — complete kit: already includes this enzyme
- S. aureus Gyrase DNA Supercoiling Assay Kit (BHT16300075) — DSA100KSE already includes this enzyme; DSA100KS is enzyme-free and needs this product
Protocol
No separate datasheet is available for this enzyme. The ProFoldin protocols “DNA Topoisomerase ATPase Assays” and “S. aureus DNA Gyrase DNA Supercoiling Assay” (linked under Documents) show how the enzyme is used in the matching assay kits.
